DMT and Spirituality: The Molecule of God, or Psychedelic Experience?
DMT produces the most intense altered state achievable by chemical means. Clinical researchers called it the "spirit molecule." Here's what 30 years of research says about those encounters.
In 1931, a chemist named Alexander Shulgin โ then working at the USDA and later at the pharmaceutical company Parke-Davis โ synthesized N,N-Dimethyltryptamine for the first time. He wasn't looking for a psychedelic. He was studying tryptamine derivatives, a class of compounds related to serotonin (5-hydroxytryptamine), and DMT was just one of many analogs. The molecule itself had been isolated from the Mexican plant Desipodera viridis in 1931 by Raphael and Glennon. Nobody knew what it did to humans for another two decades.
Shulgin eventually tried it himself, in the 1950s or 60s, well before his 1991 book PiHKAL (Phenethylamines I Have Known And Loved) made him a household name in psychedelic circles. His description of the DMT experience was unmistakable: "It's like the universe is telling you something, and you can't quite understand the language." Within a few seconds of inhalation, the physical world dissolved into geometric patterns, then into what he and later users described as an entirely different reality. The experience lasted 15โ30 minutes. The intensity was unlike any other known drug.
Rick Strassman and the clinical studies
Rick Strassman, a psychiatrist at the University of New Mexico, conducted the most systematic clinical research on DMT in the 1990s โ the first DMT studies in the United States since the 1960s. Between 1990 and 1995, he administered DMT to 40 healthy subjects in a carefully controlled setting. Each subject received 6 doses over several days, ranging from sub-perceptual to full psychedelic. The study was funded by the National Institute on Drug Abuse (NIDA) and followed rigorous clinical protocols.
The results were published in a series of papers and later compiled into his book DMT: The Spirit Molecule (2001). The most striking findings: nearly all subjects at the full dose reported encounters with "other entities" or "other worlds." These weren't subtle visual distortions. Subjects described encountering intelligent, apparently autonomous beings in what they called "machinespace," "other dimensions," or "another level of reality." Many described being greeted, examined, or communicated with by these entities. Some reported feeling their consciousness transported to an entirely different realm. A significant number described experiences they characterized as spiritual or mystical in nature.
Strassman was careful not to claim these were real encounters with other dimensions. The subjects, he noted, were healthy volunteers with no history of spiritual practice or mystical experience. Their reports, however, were remarkably consistent across individuals. The entities appeared independently of the subject's expectations. When asked what they expected before the study, most said "visual distortions" or "weird feelings" โ not autonomous intelligences.
The chemistry: tryptamines, MAOIs, and ayahuasca
DMT is a tryptamine-class compound, structurally related to serotonin. When ingested orally, it's rapidly destroyed by monoamine oxidase (MAO) enzymes in the gut, which is why it has to be smoked, injected, or combined with an MAOI to be orally active. Ayahuasca, a traditional Amazonian brew, solves this problem naturally: it contains DMT (from the Psychotria viridis vine) plus harmine, a natural MAOI (from the Banisteriopsis caapi vine). The harmine blocks the MAO enzymes, allowing the DMT to reach the brain. The combined effects last 4โ6 hours โ much longer than smoked DMT's 15โ30 minutes โ and produce a more sustained, less peaky experience.
DMT is also believed to be endogenous โ produced naturally by the body. A 2016 study by Jonathan Nash at King's College London detected DMT in human blood and urine, but at very low levels (nanograms per milliliter). These concentrations are far too low to produce psychedelic effects. The question of whether the pineal gland produces significant DMT at death (a theory popularized by Strassman and others) remains unproven. The pineal gland does produce melatonin from tryptamine precursors, but whether it synthesizes DMT at psychoactive levels is still speculative.
Modern research: the entropic brain
Robin Carhart-Harris at Imperial College London is one of the most influential modern psychedelic researchers. His 2014 "entropic brain" hypothesis, published in Neuropharmacology, proposes that psychedelics increase the brain's entropy โ the diversity and unpredictability of its neural activity. In normal consciousness, the brain operates in a highly constrained, efficient state with strong top-down control (the Default Mode Network dictating what you pay attention to). Psychedelics disrupt this constraint, allowing normally filtered information to reach consciousness. This produces the sensory overload, unexpected connections, and ego dissolution characteristic of the psychedelic experience.
Carhart-Harris's mechanism for this disruption is 5-HT2A receptor agonism. DMT, psilocybin, LSD, and mescaline all primarily act by activating serotonin 2A receptors, particularly in layer V pyramidal neurons of the prefrontal cortex. This activation appears to both increase "upward" sensory signaling and reduce "downward" predictive control, effectively removing the brain's usual filters. The brain floods with information it normally suppresses, and consciousness expands to include this noise.
DMT microdosing and the modern scene
The psychedelic renaissance has brought DMT microdosing โ taking sub-perceptual doses for cognitive enhancement or mood improvement โ into the cultural conversation. Rick Doblin, founder of MAPS (Multidisciplinary Association for Psychedelic Studies), has been advocating for psychedelic research and decriminalization since 1986. MAPS is currently running clinical trials for MDMA-assisted therapy and has funded psilocybin research. DMT research lags behind psilocybin and MDMA primarily because DMT's effects are too intense and too brief for a traditional therapeutic format. Unlike psilocybin sessions that last 4โ6 hours with manageable intensity, DMT's 15-minute explosion is hard to sit with therapeutically.
Microdosing DMT has attracted attention but minimal research. The evidence for microdosing any psychedelic is thin and inconsistent โ a 2020 review in Journal of Psychopharmacology found that most positive reports were anecdotal, and controlled studies produced mixed results. DMT microdosing is particularly untested because of DMT's short half-life and the difficulty of producing a sub-perceptual dose without producing nothing at all. The all-or-nothing dose-response curve of smoked DMT makes microdosing technically more challenging than with psilocybin, which has a longer duration and more gradual onset.
